Targeted amplification out of cDNA is what works very robustly for scRNAseq, nuc-seq recovery seems very lossy - based on experience from Ryan Delgado and Matt Keefe . Strong promoter (cag) and wpre should give you tens of UMIs per cell. All based on MOI=1 lentivirus
