We present projection imaging with parameter selection (PROPS) for light-sheet microscopy.

With PROPS, we can control the depth and location of a projection, and optically unwrap a curved surface, here of a drosophila embryo.

We present oblique plane microscopy with adaptive optics (AO):

One deformable mirror corrects both the light-sheet and the fluorescence detection.

Movie shows cancer cells in a zebrafish xenograft model, before and after application of AO.

Thrilled to present one of my works in the Prinz lab as preprint!

We find BLTP2 controls phospholipid composition and fluidity of the Plasma Membrane (PM), and tumorigenic features of cancer cells, putatively by transporting PE lipid from the ER to PM.

Our mesoscopic oblique plane microscope is online:
opg.optica.org/optica/fulltex…
It enables rapid volumetric imaging of a whole zebrafish embryo with ~2.3 micron lateral resolution.

This is enabled by merging a couple of optical innovations and tricks. 1/N

I am very excited to share my postdoc work “Morphodynamics of human early brain organoid development”,  where we take a multiscale morphodynamic view of human brain organoid development. In collaboration with Gilles Gut  Gray Camp Treutlein lab biorxiv.org/content/10.110…🧵(1/9)

We present a meso Oblique plane microscope.

A microprism below the sample tilts the sheet, improving resolution and sectioning with a low magnification objectives.

We analyzed blood flow in zebrafish embryos at 12Hz.

Blood flow in a Zebrafish embryo, imaged at 10Hz.

Imaged with Stephan Daetwyler on the meso OPM.

Downsampled as a courtesy of twitter (original video is 3200x700 pixels and 100 frames long).

Our method (Lukas Fuentes, Bewersdorf Lab, David Baddeley ) for extracting quantifiable surfaces from single-molecule localization data is now published: doi.org/10.1016/j.bpj.…! There's a single-click installer at python-microscopy.org/downloads/ if you want to give it a try.

How do you image the large and the small at the same time? We developed new 🔬 technology to image centimeter-scale specimens - including whole mouse brains 🧠 - with diffraction-limited resolution and without sectioning.

Tweetorial about Stephan Daetwyler and my attempt to forecast the future of light-sheet scopes.

Happy to pass our orbuculum along, if someone wants it.
