This is the first slide, predating the CalTRACK process, that introduced the wild idea of an energy efficiency 'meter.'

We have come a rather long way!

We presented some biology in CalTrack alongside our code. This is a short thread to dig into that data and what it might tell us about different forms of hypertrophic cardiomyopathy.

In CalTrack we found that Mavacamten has some benefit in correcting calcium transients in a hypertrophic cardiomyopathy variant of the thin filament. However, we didn't see complete resolution of the cellular phenotype.

CalTrack can be used in a variety of model systems and cell sources. Ranging primary cardiomyocytes, iPSC-CMs, organoids, and 3D tissues. You can analyse data collected using ratiometric or non-ratiometric indicators. Analysis can be performed on movies, line scans, and PMT data.

We have also used CalTrack to define changes in calcium transient in primary cardiomyocytes due to pharmacological agents.

We have demonstrated that CalTrack can also be used effectively in human iPSC-CMs to define HCM phenotypes and responses to the pharmacological agent Mavacamten.

We have used CalTrack to define changes in calcium transients observed in primary cells due to genetic variants that cause hypertrophic cardiomyopathy.
