Unexpectedly, both our reporter system and scRNA-seq data revealed a predominance of TBX5-lineage positive ventricular CMs when using a standard small molecular WNT modulation protocol for hiPSC cardiac differentiation. 5/8

To answer this, we developed a combined TBX5/MYL2 lineage tracing reporter system in hiPSCs to determine whether hiPSC cardiac diffs generate cells from the first heart field (FHF) and second heart field (SHF) lineages which give rise to the LV and RV, respectively. 3/8
