In this data-mining study, antipsychotic use in dementia was linked to all sorts of poor outcomes. But wouldn't these be inflated given the choices for controls: 'non-users using incidence density sampling' and appendicitis and cholecystitis patients?

Sjors Scheres The problematic word here is 'always'. It's mechanistically impossible for a process dominated by surface interactions (2ry nucleation & other) to sustain templating via elongation & to 'always' generate the same polymorph. That's why heterogeneity is the norm in plaques. (1/2)

Donald Gilbert⁩ presented an interesting sub analysis of the Phase 2b (D1AMOND) trial of ecopipam in Tourette syndrome. There is novel information when examining the changes in YGTSS, the main endpoint. This study has been a labor of love for Dr. Gilbert.

To be honest, I think the assertion that a specific protein fibril shape characterizes each of these complex diseases, when the fibrils don't even predict disease and lack causal mechanisms, seems a bit too reductionist.
